If they are on the same telephone line no, not at the same time.

If your WiFi signal isn't good enough to reach both rooms, then it might be worth looking at having the second router run off the first and configure it to work as a WiFi switch.
Link the two with either a long LAN cable or use a couple of HomePlug/PowerLine adapters.
